The best option from Heathrow is the Airline Bus service. The bus runs regularly from either Terminal 5 or the Central Bus Station (for Terminals 2, 3 & 4). Tickets can be bought on the bus or in advance on the website.
There are multiple stations in Oxford, and you will likely want to get off at either the Queen’s Lane or Gloucester Green stop, both of which are in downtown Oxford. The Gloucester Green stop has the benefit of a taxi rank.

Driving to Oxford is difficult due to parking constraints. Please check with your hotel if there is parking available. If not, you may want to consider a Park & Ride. There are numerous options.
